Stuffing Box Quickie - Grease

I have just repacked my stuffing box with 3 rings of PTFE Teflon material and put everything back together. For now I have just done up the nut hand tight and then locked it off. I will adjust it if need be when it goes back in the water. I did not use any grease though when packing - should I have ?
There is a grease nipple on the packing gland body just in front of the piece of hose with the clamps on at the rear where the shaft exits the boat. Should I add grease here and if so how much and what grease ?
